Collagen

supplement

Evidence tier: moderate

In plain terms

Supports joint cartilage integrity, skin elasticity, and gut lining repair, hydrolysed collagen peptides are the most bioavailable form

How it works

Plausible, not proven. Hydrolysed collagen is digested like any protein into amino acids and small peptides. A few di and tripeptide fragments (such as prolyl hydroxyproline) are absorbed whole and have been proposed to act as signals that nudge skin fibroblasts and cartilage chondrocytes to make collagen. This is a mechanistic hope supported by absorption studies, not a proven in body pathway at the doses people take.

What the evidence says

Two very different evidence pictures, and they must be kept apart. SKIN (hydration, elasticity, wrinkles) is the strongest use and grades MODERATE, but the trials are conflicted. A 2021 systematic review and meta analysis of 19 randomised trials in 1,125 people (PMID 33742704) found hydrolysed collagen beat placebo for skin hydration, elasticity and wrinkles, with about 90 days of use effective. A 2023 meta analysis of 26 trials in 1,721 people (PMID 37432180) again found significant gains in hydration and elasticity, BUT the authors say plainly that they identified several biases in the included RCTs and that further large scale randomised trials are necessary to confirm the findings. JOINTS (knee osteoarthritis) is WEAKER and grades limited: a 2018 review (PMID 29018060) found a large short term pain effect but warned the quality of evidence was VERY LOW and found no clinically important effects at medium or long term; an independent 2025 randomised trial in 68 knee osteoarthritis patients (PMID 40897777) found NO significant difference from placebo over 12 weeks; a 2026 review (PMID 42356259) concluded collagen peptides give modest, heterogeneous and formulation dependent symptom relief with no consistent evidence of structural disease modification. THE INDUSTRY FUNDING CAVEAT IS NOT A FOOTNOTE, IT IS THE POINT: the trials are small and very often industry funded. The 2025 independent trial names the problem directly, noting that earlier collagen studies were primarily industry sponsored, which raises potential bias concerns; and a 2023 trial reporting a short term pain drop (PMID 36918892) was authored partly by EMPLOYEES OF THE BEVERAGE COMPANY (Suntory) funding the product, with all groups including placebo improving similarly by 24 weeks. Also baseline honesty: hydrolysed collagen is a protein you digest into amino acids; it does not arrive at your face or your knee as intact collagen. It is not vegan (bovine, porcine, chicken or fish derived) and marine collagen is a fish allergen.

Dose and timing

2.5 to 15 g per day of hydrolysed collagen peptides, with about 10 g per day the most studied dose for skin and joint outcomes. Taken as a powder in a drink, or as capsules, sachets or ready to drink formats, with or without food. Because it is a food protein there is no meaningful drug interaction to time around.

Take 10 to 20g of hydrolysed collagen peptides daily in water, juice, or smoothies. Works best with vitamin C (which is essential for collagen synthesis). Allow 8 to 12 weeks of consistent use to assess joint and skin benefit.

Onset: ~90 days

Safety

Generally recognised as safe (GRAS). Well tolerated with no significant adverse effects reported at standard doses. Marine collagen may carry allergen risk in fish allergic individuals.

Pregnancy: caution

Breastfeeding: caution

Active compounds: hydrolysed collagen peptides (collagen hydrolysate), glycine, proline, hydroxyproline, prolyl-hydroxyproline (an absorbed dipeptide fragment)
